As the collected oral fluid (saliva) specimen travels via capillary action, drugs, and drug metabolites that may be present in the collected sample compete against their respective drug conjugates for the binding sites located on their particular antibodies. If a parent drug or drug metabolite is present in a tested sample at a level below its cut-off concentration, the substance will fail to saturate the binding sites of its particular antibody. In turn, the antibody will react with the drug-protein conjugate and lead to the development of a colored line on the test line region (T) of the specific drug strip. If a parent drug or drug metabolite is present in a tested sample at a level above its cut-off concentration, on the other hand, all the binding sites of the antibody will be saturated, and a colored line will appear in the test line region (T). In addition to the test line (T), a line should appear in the control region (C) to serve as procedural control and confirm the validity of the results.

Before starting the procedure, it should be ensured that each component of the kit is brought to room temperature. The tested person must avoid consuming food, drink, gum, and tobacco products or placing anything in their mouth for at least 15 to 20 minutes before sample collection. To collect an oral fluid sample, the provided collection swab and the collection tube are removed from their sealed pouch. Next, the collection swab is inserted into the mouth, the cheeks, gums, and tongue are swept with the help of the swab, and the swab is held in the mouth for a few minutes until the saturation indicator becomes present in the indicator window of the collection swab. Then, the collection swab is removed from the mouth and pushed headfirst into the screening device until it locks in place. Finally, the device is kept upright on a flat surface, a timer is started, and results are read in the reaction time specified in the manual.
